Oncogenic role of MiR-130a in oral squamous cell carcinoma

Abstract Aberrant activation of the PI3K/AKT/mTOR pathway is attributed to pathogenesis oral squamous cell carcinoma (OSCC). In recent years, increasing evidence suggests involvement microRNAs (miRNAs) in carcinogenesis by acting as tumor suppressors or oncogenes. TSC1, a component above pathway, regulates several cellular functions such proliferation, apoptosis, migration and invasion. Downregulation TSC1 reported well other cancers associated with an unfavourable clinical outcome patients. Here we show that oncogenic miR-130a binds 3′UTR represses its expression. MiR-130a-mediated repression increases anchorage independent growth invasion OSCC cells, which dependent on presence TSC1. We observe inverse correlation between expression levels samples, suggesting their interaction physiologically relevant. Delivery antagomiR-130a cells results significant decrease xenograft size. Taken together, findings study indicate miR-130a-mediated downregulation not only novel mechanism OSCC, but also restoration may be potential therapeutic strategy for treatment OSCC.
